iGEM EPFL 2018
We received the plasmid from the repository in BioBrick format and successfully amplified it using PCR. We inserted the part using Golden Gate assembly (BsaI) into a pet14 vector under a T7 promoter and terminator and confirmed the assembly using Sanger sequencing. We expressed the construct in TX-TL cell-free medium, but were unfortunately unable to determine for sure whether expression occurred and the 60-mer was formed as bands were not clear on SDS PAGE gels.
